Chain initiation in a polycistronic message: sequential versus simultaneous derepression of the enzymes for histidine biosynthesis in Salmonella typhimurium.

چکیده
The pathway for histidine biosynthesis in Salmonella typhimurium, elucidated largely through the efforts of Dr. B. N. Ames and co-workers,1-5 is shown in Figure 1. The figure also shows that portion of the pathway for purine biosynthesis which involves the further metabolism of 4-amino-5-imidazole carboxamide ribonucleotide (phosphoribosyl-AIC).6 Dr. Phillip Hartman and his colleagues have shown that the structural genes for the enzymes catalyzing the reactions of the histidine pathway are localized in a small segment of the bacterial chromosome, the histidine operon.7' 8 The phenomenon of coordinate derepression of the histidine enzymes was originally described by Ames and Garry,9 who found that the histidine operon functions as a single unit in response to the level of histidine available to the organism. Evidence presented by Martin10 indicates that the histidine operon is transcribed into a polycistronic messenger-RNA, and studies on polarity in the histidine operon7' 11,12 suggest that this messenger-RNA is translated from the end which

Histidine regulation in Salmonella typhimurium. XV. Procedure for the selection of mutants unable to derepress the histidine operon.
A general search has been made for mutants defective in their ability to derepress the histidine operon. The procedure was to select for mutants with an increased sensitivity to the false feedback inhibitor, 2-thiazolealanine. Five mutant strains defective in derepression have been isolated. All five strains are unable to derepress normally because of mutations located in the operator-promoter ...
متن کاملFirst enzyme of histidine biosynthesis and repression control of histidyl-transfer ribonucleic acid synthetase of Salmonella typhimurium.
The regulation of formation of histidyl-transfer ribonucleic acid (tRNA) synthetase was examined in strains of Salmonella typhimurium. When the first of the histidine-forming enzymes was wild type, the presence of 2-thiazolealanine in the growth medium prevented repression of histidyl-tRNA synthetase formation elicited by the addition of 1, 2, 4-triazole-3-alanine to these cultures. Conversely,...
متن کاملRepression of the histidine operon: effect of the first enzyme on the kinetics of repression.
Kinetic studies on repression of the enzymes for histidine biosynthesis in Salmonella typhimurium showed that, upon addition of histidine to a derepressed culture, the enzymes became repressed in a temporal sequence which corresponds with the positional sequence of the genes in the histidine operon. This serial pattern of repression occurred under conditions in which the feedback site of the fi...
متن کاملControl of isoleucine, valine, and leucine biosynthesis. VI. Effect of 5',5',5'-trifluoroleucine on repression in Salmonella typhimurium.
The leucine analogue 5',5',5',-trifluoroleucine (fluoroleucine) replaced leucine for repression of the isoleucine-valine biosynthetic enzymes in Salmonella typhimurium. In contrast, the analogue had no effect on derepression of the leucine biosynthetic enzymes in leucine auxotrophs grown on limiting amounts of leucine. The effect of fluoroleucine on repression appeared to be specific for leucin...
متن کاملHistidine regulation in Salmonella typhimurium. 8. Mutations of the hisT gene.
The hisT gene, one of six genes in which mutation causes derepression of the histidine operon in Salmonella typhimurium, is shown to code for a protein that is not essential for the growth of the bacteria. This is indicated by the characterization of particular classes of mutations in the hisT gene: amber mutations, frame-shift mutations, and temperature-sensitive mutations that affect repressi...
